Interests

Mycobacterium tuberculosis (M. tb) is a dreadful pathogen causing about 2 lakh deaths a year. The heat shock proteins of this organism are reported to be antigenic and also are able to elicit immune response, as many of them are secretary. The Hsp70 of Mycobacterium is reported interacts with the extracytopalsmic domain of CD40 and elicits chemokine release. Expression of this is regulated by a co-repressor HspR. Having the importance of mankind we are interested to study the molecular aspects of some of these proteins for better understanding the biology of the organism. We
would like to study the functional aspects by biochemical and biophysical characterization of some of these proteins as well as the complexes they make.

My work involves the following lines of interests:

Structurally and functionally analyze Hsp70 and the complexes of Hsp70 with HspR and hCD40.

Purification and characterization of hCD40 using Pichia pastoris Expression System.

Generation and characterization of the different swapped mutants that complement the loss of functional E. coli groEL and/or dnaK.
