Caffeine-driven thoughts of a forest ecologist

    • Good primer, bad primer

Further to my post on good samples and bad samples, I am happy to report that there was enough DNA in the bad samples and that our primers work (although some are not specific enough).

Good primers, earlier today (slightly enhanced in PS to make this post look more dramatic)

Cool, are all the bands from the coffee samples?
          how much DNA was there?
          that’s inspiring, actually.
          …what gene were u amplifying?

We tested 20 primer pairs (EMBL numbers on top) on 8 coffee samples (codes at the bottom). DNA concentrations were very low (avg 11-20 ng/ul). We were amplifying SSR primers. You can follow everything on my Open Notebook.
