Hi
I am trying to release a nuclear protein from my cells in order to do a western blot. I was intending to use NP-40 but have been told it may not penetrate the nuclear envelope. Does anybody have any suggestions as to what works well in this situation and to whether NP-40 can be of any use?

Bio-Rad has a nuclear prep kit (full disclosure – I work for them) as does Pierce (NePER) and there are likely others. These are multi-step separation kits, but work well in general.
