  Isolation of PBMC can be achieved by simple centrifugation isolation, there after you can culture them to get monocytes/macrophages.but this method will activate monocytes. If you want to isolate pure monocytes derectly fom blood or from PBMC, then easiest way is to go with monocyte specific CD14+ beads. you can fine information from the link below

  Hi Pooja,
  I do not know whether this still helps you, but I also had trouble with monocytes in 10% FBS and RPMI 1640. The problem was the FBS. We changed the company and everything was fine.
