Request for a protocol for extracting RNA from cells in agar.

Great Forum!!! I was wondering if anyone has tried extracting RNA from cells in Agar. I was growing some cells to look at the colonogenicity and wanted to look at the message level also but I cannot find a way to extract RNA. Qiagen tech support was not able to help. If anyone has any suggestions please let me know!!!

    • I would suggest using the Bio 101 “FastPrep” method: It’s an extraction kit that uses a small tube half filled with small beads which is shaken vigorously in a specialized machine (known as a bead-beader or “FastPrep machine”). The tube contains chemicals that stabilized the DNA/RNA as it is released from the lysed cells.

      I haven’t worked with cells in agar my-self, but I have successfully used this method for extracting DNA (an RNA kit is also available) from really difficult sources like raw soil samples.
