Dear friends,
I am trying to do Tunel Test (apoptosis assay) on adherent cells(SHEP cels) but always I find a high background …actually all my cells turn to green…I hope you have any sugestion!
I use In situ cell death detection kit, fluorescin from Roche

I don’t remember exactly what’s in the kit, but can you do a negative control without the active component (terminal transferase?) and with the fluorescin? This should label nothing. If it does, try lowering the concentration of the fluorescin until you don’t see background anymore, and then retry everything with the active component.
