how to do imaging on isolated myocytes?

Hi
I’m currently working with fluorescence imaging on isolated skeletal myocytes. i need to image them immediately after isolation. they are rod shape cells,very thick in nature and will be floating in the medium. to permeabilize the cells i’m using saponin (50 microgram/ml).
can anyone tell me how much maximum concentration saponin i can use for permeabilization and incubation time.
is there any specific method to image the isolated skeletal myocytes.
i use fluo4, rhod2 and ER tracker blue fluorescence probes.
