Hi Madhura,
Thank you for using the Nature Protocols Discussion Forum.
Can I ask if you are looking for a general method for isolating and fusing protoplasts, or would you prefer methods specific to a particular species?
Thanks,

Hello Dorothy,
Thank You for posting a reply.
Well, I am in search for a protocol for isolating and fusing protplasts using
1.yeast cells &
2.plant cellsThanks.

Dear Madhura,
I’ve searched our published content and didn’t find anything relevant to you on the Protocols Network (our freely available content). We do have a few Nature Protocols articles (access to these requires a site license) that describe plant protoplast isolation for a number of applications – but not for fusion, and not for yeast. I’ve added links to the abstracts of these below; perhaps the authors will be able to help you out?
Arabidopsis mesophyll protoplasts: a versatile cell system for transient gene expression analysis
Transient expression of fluorescent fusion proteins in protoplasts of suspension cultured cells
Isolation of intact vacuoles from Arabidopsis rosette leaf–derived protoplasts
Our content is updated weekly so it’s worth searching regularly for new articles that may be more directly relevant.
Good luck with your experiments!
