Lamina Propria Lymphocyte isolation

I am adapting the Nature Protocols protocol for the isolation of murine lamina propria mononuclear cells. The protocol calls for use of 5ug/ml of amphotericin B in the culture media following isolation. This is twice the suggested concentration, so I have two questions. Can a lower concentration of amphotericin be used and still be protective? What does the amphotericin at that concentration do to the cells in culture?

Hi all,
As this question concerns one of our Nature Protocols articles please could you post any discussion to http://www.natureprotocols.com/2007/09/20/isolation_and_subsequent_analy.php ? This will collect all the comments together in one place and enable users who find the protocol via PubMed or our site to have the benefit of your feedback as well.
For the next month you can access the protocol itself for free here .
Please note you can post questions and comments about all our protocols via the abstract, we’re hoping this will keep our protocols up-to-date.
Thanks,
Katharine
