Reply from Rasmus Wernersson

I would suggest using the Bio 101 “FastPrep” method: It’s an extraction kit that uses a small tube half filled with small beads which is shaken vigorously in a specialized machine (known as a bead-beader or “FastPrep machine”). The tube contains chemicals that stabilized the DNA/RNA as it is released from the lysed cells.

I haven’t worked with cells in agar my-self, but I have successfully used this method for extracting DNA (an RNA kit is also available) from really difficult sources like raw soil samples.

Link: http://www.qbiogene.com/fastprep/
(It appears that Bio 101 is now part of Q-Bio Gene).

I hope this can be used
-Rasmus Wernersson
