Dear suppe,
You can measure the activity with to different methods. One of them use a Clarke-type oxygen electrode to obtain the respiratory activity of the ETC (or ETs). The other way is to measure the change of absorbtion qhen it works. I recommend you this work:
J.B. Clark, T.E. Bates, P. Boakey, A. Kuimov and J.M. Land, Investigation of mitochondrial defects in brain and skeletal muscle. In: A.J. Turner and H.S. Bachelard, Editors, Neurochemistry: A Practical Approach, Oxford University Press Inc., New York (1997), pp. 151–174.
