Proteasome inhibitors and Cytotoxicity

Hi everybody,
I am working on proteasome inhibitors (i.e., Velcade) and cellular toxicity and right now I am looking for protocols in order to induce stress to cells, any stress! Any hints?

    • In our Lab, we work on Cancer cell lines. Usually we use G418 based cytotoxic stress.

      Maybe I should have been more clear. I work with Multiple Myeloma cells and I need to find out newer ways than antibiotics in order to induce cytotoxicity. The problem is my ignorance in molecular biology protocols because I am formed as a Neuroscientist! =(

      For example I would like to induce:
      Oxidative stress
      Mitochondrial stress
      ER stress
      Lysosomal stress

      How can I?!?

    • Hi Luca,

      Well, simvastatin can be used to induce mitochondrial stress by inducing caspase pathway. I came across a paper that reported synergistic induction of cell death by the combination of simvastatin and doxorubicin. (Check this: Br J Pharmacol. 2004 November; 143(6): 715–724)

      It looks like it induces apoptosis by activation of mitochondrial caspase 9 in lymphoma and myeloma cells: that’s perfect.
      Now I just have to look for the others stressors…
