changing peak shapes

I am in the process of developing a HPLC assay for the estimation of clobazam.In the first two std curves, the shape of clobazam peak was fine. But on the 3rd std curve, the shape of the clobazam peak changed drastically.It appeared to come with the previous peak of N desmethylclobazam and quantification was impossible.this happened for the lower concentration- 10-100ng/ml of clobazam.

Can any one please tell em why this happened?

      Many times things work well once or twice, but not in the final analysis. I think this is a case in point.

      I think your mobile phase B may have run out, but it’s just a suggestion.

      If one examines ones relationships, you may find a reason for this… and a way to move forward.
