MS of difficult proteins (0 replies)

Could anyone provide some advice on analysis of difficult (large >50kDa and contaminated) proteins using MS? Advantages and disadvantages of using ESI-MS and MALDI-MS for this purpose? Is the concentration of 1 mg ml-1 high enough to see anything?
Thank you in advance for your comments.
P.
